Genetic Fragment Analysis
We perform capillary electrophoresis size separation of your fluorescent-labelled fragments on our Applied Biosystems 3730xl Genetic Analyzer.
The CHGI accepts fluorescent dye labelled fragments for size separation using capillary electrophoresis only. The upstream sample manipulations (labelling, amplification, digestion, etc.) and downstream analysis of the data are the responsibility of the customer.
As of October 3, 2022, results will be returned every Monday, Tuesday, and Wednesday.
Results are posted on our secure web server. Fragment analysis files are available in .fsa format.
If you have questions or concerns, please don't hesitate to contact us. We are here to help!
General Guidelines
Please submit at least 5 μL of each samples.
Considerations:
- You may have to dilute your labelled PCR products in order to determine the optimal amount for loading. Try serial dilutions of samples before submitting a large project.
- You may have to perform a desalting step on your PCR products.
- Samples can be multiplexed to increase throughput. Keep the following in mind:
- Use different dye labels for samples with overlapping product sizes.
- Use an acceptable dye combination (see table below).
Acceptable Dye Combinations
Dyes and Size Standard | Filter Set |
---|---|
6-FAM, VIC, NED, PET with LIZ Size Standard | Filter set G5 |
6-FAM, HEX, NED with ROX Size Standard | Filter set D |
5-FAM, JOE, NED with ROX Size Standard | Filter set F |
Available Size Standards
We will select the appropriate size standard from the table below based on the filter set selected and size range of the fragments recorded on the order request. If you require any other size standard for your analysis, please provide the standard with your samples.
Size Standard | Maximum Fragment Sized (bp) |
---|---|
GS 500 ROX Size Standard | 500 |
GS 500 LIZ Size Standard | 500 |
GS 1200 LIZ Size Standard | 1200 |
